I don't know if you can sign on in Porto, but a few years ago we took a 5 day (7 day from Lisbon) cruise on the Uniworld line from Porto to the end of the navigable part of the Duoro, then the cruise included a 1- day trip to Salamanca ( a highlight) and back to the ship and Porto. We loved it. Unlike some of the other river cruises we have been on, this one doesn't travel at night, so you see the full river panorama in daylight.
Viking offers decent cruises up the Douro from Porto (http://www.vikingrivercruises.com/regions/europe/rivers/douro/index.aspx), but if you would like to really enjoy the area, you might want to consider either staying at one of the excellent Quintas or rent a property overlooking the river. It would be much more enjoyable.
