Is this a difficult trip to make in one day? Is this an "easy" drive (multiple lane highways) or "hard" (single lane, windy roads)?
Where would you stay over night on the way from Amboise to St Remy? Avoid cities if possible!

Yes, of course you can take multiple-lane highways (that are far from "easy" IMO) just about all the way, but IME it's not the slightest bit interesting or fun. I'd get myself to Avignon on the train, then pick up a car for an easy drive to St-Rémy.
It is almost "all freeway" from Amboise to St Remy. www.Viamichelin.com says it is 7 hr 10 mins - but I would count on a minimum of 8 hrs for "deviations", pottie breaks, meals etc. I've driven about 80% of that route several times & it is not very scenic until you get to upper Provence.
Consider a train
- Amboise to Avignon TGV. Leave at 1:57 & arrive at 6:45 - 1 train change at St Pierre des Corps
- St Pierre des Corps (near Tours) to Avignon TGV.
. Leave 10:00 arrive 2:38 1 train change in Marne la Vallee (don't go into Paris for a train change)
. Leave 2:31 arrive 6:45 no train changes.
It is a 30-45 min drive from the Avignon TGV station (plenty of rental cars there) to St Remy.
A TGV trip is VERY relaxing, IMO.

Verify the train schedule for your specific travel day. Often there are different schedules on weekends. I don't know if the summer schedule has been released yet. I picked a Tuesday in April.
