How to best spend a day there? What not to miss? Suggestions for not having to stand in lines at entrance. Suggestions for getting tickets for the boat ride. Is taking the bus from Leiden better than taking a car to the gardens? Any other suggestions and ideas would be great!

It is kinda hard to "miss" anything if you stay long enough and if nothing else you'll see acres of flowers depending on when you go (I last went in April and it was perfect for me).
Took a bus from Schiphol Airport (I was staying in Amsterdam and took the train out to the airport and then caught the bus). We certainly didn't stand in any lines at the entrance but we also got there sometime after it had opened for the day.
The place was packed with people but that didn't, somehow, detract in any way from the viewing.
Best way to go? Go Dutch - bike it. http://tinyurl.com/y8hltjl. Bring a bike lock and chain.
The tulips fold up at night so go on a warm sunny afternoon for a good view.
