No, no, and yes. The bus does not leave the main highway.
The TicaBus does not go through Monteverde. As Jeff said, it does not leave the main highway, the InterAmerican or Route 1. They always take the same route with no stops except for Liberia.
However, you can ask the drive to drop you off at one of the entrances to Monteverde and catch the bus from there to Monteverde. (Check your map.) A convenient stop would be at Cuatro Cruces.
But being let off on the highway, your timing would have to be impeccable. There are very few public buses that go to Montevederde period. You could be waiting a long time.

All you have to do is check www.thebusschedule.com for buses to Monteverde from San Jose or from Puntarenas. They both pass by this spot. Then you can schedule your TicaBus accordingly. There are some sodas and small shops in that area so you can get something to eat or drink while you are waiting.
