If you are a Costco member (or decide to join) there is one on Hwy 111. Probably your best bet.
After that I would probably say Henry's on Hwy111,
Trader Joe's is also in La Quinta, and it's a novelty if you don't have them at home. They started as an outlet for wine and snacks, now they carry a full range although I wouldn't say their produce and seafood is necessarily the best quality nor the freshest, but it's convenient.
Just get to Washington and hiway 111 and a number of grocery stores, etc. There is also a Stater Bros - that sometimes has great buys, such as one Thanksgiving, they had leftover Prime - repeat - Prime - Ribeye roasts - for $1.99 a lb - and they even cut them up for us.
Trader Joe's has good frozen fish and other frozen entrees, smoked salmon, salami, etc - a wide selection of cheese, etc.
And I think there is also a Sprouts (formerly Henry's) at the shopping center by the intersection (NE corner - or a few blocks east on hiway 111) and they have fresh olives, spices by the ounce, a good meat counter, whatnot.
Between the two of them, you can pretty much find what you want.

As noted, Henry's is now Sprouts. Good suggestions on Costco and Trader Joe's.
There is a Fresh N' Easy on Calle Tampico West of Washington Street for a wide selection of fresh produce and prepared foods.
I'm surprised no one picked it up, but one of the best venues for reasonable, fresh produce, meats, cheese, baked goods, etc is the Certified Organic Farmer's Market every Sunday morning at Old Town La Quinta. Highly recommended.
Also should mention La Quinta Wine Co on Washington near Avenue 47. The owner, Greg, relocated from the Bay Area after operating a prominent wine store for many years and has a wide selection of wines at a range of price points. If you're like me, you will look for "the $25 cab that drinks like a $75 bottle." He will steer you right.
Besides that, Trader Joe's can be a good resource for some bargains (but only if you know what you're looking for), and Costco does a good job with that too.
One of the best things I have found there is Summerland Wolff Creek Pinot Noir for $20/bottle, easily $30 at other stores.
