My husband & brother are driving from East Coast Canada to Boston where they are picking me up at the airport. From Boston we are planning a 5 - 6 day bike ride to the Vernmont area. As we are not familiar with this area we would love some suggestions on what routes to take and hotels and anything else that you can offer.

On a bike, you might want to take MA route 2 west from Boston to rt.7 north to VT and then loop back east through NH to ME then south to Boston. Of course you could loop around the other way. Rt. 2 will take you to Concord and Lexington for some history, a short way outside of Boston.
Good suggestion above to take Route 2 out of Boston, but I would then take US 202 from Gardner, MA, to Jaffrey, NH, and explore the Monadnock Region, including Peterborough. Then take NH and VT 9 across to Bennington, home of Hemmings Motor News. Head up 7 from there toward Burlington. When you come back, take NH 100 an incredibly scenic ride, then east on US 4 to Woodstock, VT. From there, you continue on US 4 into NH and can turn north to tour the White Mountains when you reach Interstate 93. You don't say where you are leaving from, but you can get a bus back to Boston from several points in NH or you can stay on the bikes and go back to wherever in Atlantic Canada.
