You can fly to Anegada on a charter flight (affordable if you are traveling with a group), but all the other islands are reachable only by boat (or perhaps helicopter if you are visiting Necker Island). Anegada has ferry service from Tortola (via Virgin Gorda) three days a week, while Jost Van Dyke has regularly scheduled ferry service from Tortola, St. John, and St. Thomas, but not all these services run daily. You can rent a jeep on Anegada, and many people do, especially if they are staying for several days. It's possible to rent a car or jeep in Jost Van Dyke, but it's not really necessary.
