I assume you are self driving? If so it is a good idea to split the journey into north and south with a stop in Salta. Check out Casa Hernandez a great B&B in San Lorenzo just outsid the city. Alex and Reijke are terrific hosts.
We made the mistake of getting to the Salar via San Martin de las Cobres, a god forsaken place if ever there was one and terrible roads!
Tilcara has arguably a better choice of accomodation but we found it a bit too toursisty. I would try to make sure that you stay one night in Purmamarca so you can photograph the hills in the best light which is first thing in the morning.
There is a link to my trip report of our last visit @ http://www.fodors.com/community/sout...th-america.cfm scroll down to Oct 1 for the relevant section. There is some stuff re te vineyards we visited in Cafayate which might be useful. There is an earlier TR from 2008 but good luck finding that!
I would definitely suggest adding Cachi for a night en route to Cafayate. A very pretty town with a intersting hill top cemetery. You could also then visit Molinos on your way down Ruta 40 . It has one of the prettiest churches in South America and an amazing, if expensive hotel in what is a tiny village.
