How long will you be gone? What's your destination in Wyoming? I'm assuming it's Yellowstone and/or Grand Tetons.
On the way out, I'd definitely stop at Mount Rushmore in SD. (I don't have any suggestions between PA and there although I'm sure others will).
Be sure to stop in Cody. Keystone SD to Cody WY is about 6 hours (according to Google Maps).
If you have lots of time, I'd detour south and head towards Denver. If you are going in July, stop in Cheyenne for the big rodeo - Frontier Days.
If you want more mountains, Rocky Mountain National Park is great (Estes Park, CO). South of Denver is Cripple Creek and it's fun to go down in a gold mine - The Mollie Kathleen mine. You may want to go up to Pike's Peak too.
Head east along the plains to St. Louis, MO and go up in the Gateway Arch.
They would probably enjoy Mammoth Cave at Lincoln, KY.
On the way home, you could stop in Beckley WV and visit a coal mine. It's run by the city of Beckley and is a cool experience.
That route is 4417 miles and 3 days, 2 hours.
I wouldn't drive more than 500 miles a day so that's 9 days of driving. Make sure you have at least that many days of "being" somewhere and experiencing stuff along the way.
Those are things I think two boys 8 and 10 may enjoy.

Depending on what part of PA you are coming from this place could be your 2nd or third nights campground- http://www.iowadnr.gov/parks/state_p...wis_clark.html
Use I-80+I-680 across Iowa to get on I-29 along the Missouri River. The next day stop at Mitchell SD to see the "Corn Palace"
Before you get to Rapid City make a brief detour into the small town of Wall to see Wall Drug. You will see signs for it for hundreds of miles. Camp somewhere near Rapid City. Next day see Mount Rushmore and the Crazy Horse Memorial. There is plenty of other stuff to see in that area if you have the time. In the NE corner of Wyoming detour off I-90 to see Devils Tower. Camp somewhere near Cody off US 14 before going into Yellowstone.
Unless you want to make a detour through Cincinnati, Kentucky and Beckley WV are a bit out of your way.

Possible stop on the way to Iowa: http://www.go-camping.com/site/index.asp Near South Bend Indiana.
Depends on what part of PA you are coming from.
