The M60 will take you through Harlem to the Columbia Univ. area. Once in Manhattan you can connect to just about anywhere by subway.
Easiest is a taxi, and will run you about $40-45 all totaled.

It depends on where you're going, how much your time is worth and how many you are. There's an Express Bus for $12/person that's good if you're staying near Grand Central or Port Authority and traveling alone. http://www.nyairportservice.com/ Once there are 2 of you, I think it makes more sense to get a taxi or order a car service for a fixed rate. dial7.com The website hopstop.com will give you all the options.

Assuming your destination is on the island of Manhattan, a taxi or car service is your best means of transportation (unless you are truly destitute AND time means nothing to you).
LGA is poorly served by public options.
